Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature. A suggested positive control is skin tissue.
c-FOS is a nuclear phosphoprotein which forms a tight but non-covalently linked complex with the JUN/AP-1 transcription factor. It has a critical function in regulating the development of cells destined to form and maintain the skeleton, and is thought to have an important role in signal transduction, cell proliferation and differentiation.
IP-MS enrichment of FOS (Intensity): FOS was enriched 1610-fold from SKMEL5 lysate compared to background proteins, using the optimized IP-MS workflow with Pierce MS-Compatible Magnetic IP Kit protein A/G (Part No. 90409) and c-FOS antibody (Part No. PA1-37437).
