Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature. A suggested positive control is placenta, lung or ovarian carcinoma.
NOS oxidized a guanidine nitrogen of arginine releasing nitric oxide in the form of a free radical and citrulline. Nitric oxide thus generated acts as a messnger in diverse functions including vasodilation neurotransmission, anti-tumor and anti-pathogenic activiteis. NOS is classified under three types: neuronal NOS (nNOS) or brain (bNOS); induciblr NOS (iNOS) or macrophage NOS (mNOS); and endothelial NOS (eNOS). This antibody reacts with eNOS.
