For dilution, use a buffer containing carrier protein, e.g. PBS with 1% BSA or a neutral media containing 10% FCS and freeze in aliquots at -70°C.
Activity: The biological activity of recombinant equine IL-8 was determined by its ability to induce equine neutrophil migration in vitro. The ED50 (or 1 unit of activity) for this effect in our laboratories is typically 22 ng/ml.
Expression: Mature equine IL-8 was expressed in Pichia pastoris, X-33. GenBank® Accession # AAO37764,
amino acids 23-101. For protein verification, the expressed protein was N-terminal sequenced and begins with the following 10 amino acids: *ITAELRCQCI. *The first five amino acids of the expressed protein appear to be cleaved from the N-terminus.
Reconstitute with 0.5 ml of distilled water.
The protein encoded by this gene is a member of the CXC chemokine family. This chemokine is one of the major mediators of the inflammatory response. This chemokine is secreted by several cell types. It functions as a chemoattractant, and is also a potent angiogenic factor. This gene is believed to play a role in the pathogenesis of bronchiolitis, a common respiratory tract disease caused by viral infection. This gene and other ten members of the CXC chemokine gene family form a chemokine gene cluster in a region mapped to chromosome 4q.
