This antibody is predicted to react with mouse and rat based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is breast carcinoma or mantle cell lymphoma.
Cyclin D1, otherwise known as PRAD-1 or bcl-1, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms, including mantle cell lymphomas (MCL). It is known as a key cell cycle regulator and functions in association with cdk4 and / or cdk6 by phosphorylating the Rb protein.
Cyclins are proteins that govern transitions through distinct phases of the cell cycle by regulating the activity of the cyclin dependent kinases. In mid to late G1, Cyclin D1 shows a maximum expression following growth factor stimulation. Cyclin D1 has been successfully employed and is a promising tool for further studies in both cell cycle biology and cancer associated abnormalities.
