Simple-to-use solutions for dengue, chikungunya, and Zika virus detection: just add RNA

Zika virus is a single-stranded RNA arbovirus that is spread to humans primarily through the bite of an infected mosquito of the Aedes genus, causing Zika virus disease. Infected individuals may have fever, rash, joint pain, and/or conjunctivitis. Studies have shown that Zika virus can be passed from an infected pregnant woman to her unborn baby, and may be linked to birth defects.

Detection of Zika and other arboviruses

Although chikungunya, dengue, and Zika viruses can be detected by testing serum for the presence of viral nucleic acid or virus-specific immunoglobulin M (IgM) and neutralizing antibodies, serological cross-reactivity is strong between Zika and dengue viruses. Molecular detection using real-time PCR is strongly recommended.

See the CDC’s updated information on diagnostic testing for Zika, chikungunya, and dengue viruses in US public health laboratories.

Real-time PCR detection of Zika and arboviruses

Thermo Fisher Scientific offers three types of Zika and arbovirus real-time PCR solutions. Each of these kits offers the following:

  • Less than five minutes hands-on time
  • Easy to use–simply add RNA
  • No cold-chain management required; ship and store at room temperature
  • Lyophilized with one-step qRT-PCR master mix
  • TaqPath Zika Kit (EUA) optimized for the Applied Biosystems QuantStudio Dx Real-time PCR system
  • All other RUO TaqMan Zika and Arbovirus kits optimized for the Applied Biosystems QuantStudio 5 Real-Time PCR System; also suitable for the 7500, 7500 Fast, and other QuantStudio systems

The Zika virus sequence used to develop these primers and probes was identified in a 2014 research paper by Pyke et al., titled “Imported Zika Virus Infection from the Cook Islands into Australia, 2014” (view full paper).

For viral RNA purification, we recommend use of the Applied Biosystems MagMAX Pathogen RNA/DNA Kit.
