When Worlds Converge™: A New Approach to Analyzing CE and MPS STR Data

This presentation will provide insight into the utility and capabilities of the new Converge software when analyzing difficult forensic samples such as bone, teeth, and highly decomposed human remains. Data will be presented to demonstrate how the features of Converge can help provide additional and more probative genetic information from a range of environmentally challenged and aged skeletal samples.

Skeletal samples were genotyped using both traditional capillary electrophoresis (CE) with the GlobalFiler DNA Amplification kit, and massive parallel sequencing (MPS) with the Precision ID GlobalFiler Mixture ID Panel. Library preparation, templating and chip loading was performed on the Ion Chef System and then sequenced on the Ion S5 System with an Ion 530 Chip. The Converge post genotype sample analysis tools were used to analyze the sequencing data, and combine the CE and MPS data for a more effective inspection of results on a case-by-case basis.

Data will be presented that compares the genotyping success when both CE- and MPS-based STR analysis is performed on challenging skeletal samples. This presentation will also discuss the additional genetic information that may be obtained when using the Precision ID GlobalFiler Mixture ID Panel in combination with the Converge software.
