Description

The EVOS® Onstage Incubator is an environmental chamber designed specifically for the EVOS® FL Auto Imaging System. It enables precise control of temperature, humidity, and three gases for time-lapse imaging of live cells under both physiological and non-physiological conditions. Environmental settings and image acquisition parameters are all seamlessly integrated into the EVOS® FL Auto interface, creating a high-performance inverted imaging system with excellent flexibility, ease of use, and a superb optical performance for demanding live cell time-lapse imaging experiments.

Fully Integrated Environmental Control System
The EVOS® Onstage Incubator affords a complete solution for environmental control during live cell imaging studies. Designed for use with the EVOS® FL Auto Imaging System, the EVOS® Onstage Incubator consists of an environmental control unit and an environmental chamber that is precision-tooled for the automated X-Y stage. A small, separate control unit supplies the power, gas (air or air-CO2 premix, CO2, and N2 for O2 displacement in hypoxia experiments), and control of humidity and temperature. Unlike many other systems that require separate software modules or control units, the EVOS® Onstage Incubator is operated through the software of the EVOS® FL Auto imager. Following a simple installation, the incubator is automatically recognized by the imaging software and is ready for use.

Seamless Operation
The EVOS® Onstage Incubator is controlled by the software and user-interface on the EVOS® FL Auto Imaging System. This creates a fully-integrated, digital, LED-based inverted imaging system with full environmental control, automated image acquisition, precision stage movement, and a streamlined user interface to deliver unprecedented ease-of-use in time-lapse imaging studies. The system is exceptionally configurable through our broad choice of high-quality LWD and oil-immersion objectives from 1.25X to 100X, proprietary LED light cubes, and vessel holders that accommodate multiwell plates, T-25 flasks and Petri dishes. The fully motorized X-Y stage and automated objective and LED cube changes helps ensure precision alignment during scanning and time-lapse experiments. Advanced image acquisition features, which include the ability to set 96 beacons for multi-position scanning, z-stacking options, and metadata that includes environmental parameters, makes the system superbly capable to meet the most demanding imaging requirements.

Flexible and Precise Environmental Control
When performing live cell imaging studies, such as cell division, differentiation, migration, and membrane and organelle dynamics, it is essential that the physiological and biological processes under investigation are not altered in any way. Shifts in temperature of even few degrees can have profound effects on dynamic cellular events, as can changes in concentration of CO2 or oxygen, and humidity levels. The ability to maintain temperature control with a precision of 0.1°C helps maintain cell viability and function and reduces the risk of biological temperature-related artifacts. It also minimizes thermal contractions or expansions that may cause focus instability or drift; the EVOS® FL Auto autofocus system helps ensure that focus is maintained even during extended time-lapse studies, when cellular movement may shift the focus plane. To enable studies of cells under normal, as well as hypoxic, conditions the system has three gas line ports. By using N2 gas it is possible to simulate hypoxic conditions by setting the oxygen sensor to the desired level.

Designed for Live Cell Imaging
Many cells are highly sensitive to light and it is important to minimize light exposure that can cause photo-toxicity, particularly over the course of an extended time-lapse experiment; the digitally controlled light output from the proprietary EVOS® light cubes enables light exposure to be minimized. The optical and detector components of the EVOS® FL Auto system, from the sensitive monochrome and color cameras to the high-quality objectives, maximize light collection. Further, the heated lid of the EVOS® Onstage Incubator helps prevent condensation and a glass coating of indium tin oxide maximizes light transmission. By optimizing illumination intensity, the number of light channels, z-steps, and image acquisition intervals, it is possible to dramatically reduce photobleaching and phototoxic effects.

User-centered Design
A user-centered design is a hallmark of all EVOS® instruments and similarly drove the EVOS® Onstage Incubator development. The control unit has a small footprint that makes it easy to move as needed and also helps save valuable bench space. The EVOS® FL Auto Imaging System and the EVOS® Onstage Incubator operate as one fully integrated unit, for an exceptional live-cell imaging workflow experience. Like other EVOS® systems the Onstage Incubator was designed to maximize performance and optimize workflow.
