The Thermo Scientific DRAQ5™ Fluorescent Probe is a far-red DNA stain for live or fixed cells and easy multiplexing with other fluorophores, such as cells expressing green fluorescent protein (GFP) fusion proteins.

DRAQ5™ Fluorescent Probe is a far-red DNA stain for fluorescent cellular imaging applications with live or fixed cells. Because of its far-red excitation and emission, the DRAQ5 Stain can be multiplexed with many other fluorophores. It is commonly chosen to counterstain nuclei and measure DNA content in cell proliferation studies involving cells expressing green fluorescent protein (GFP) fusion proteins. DRAQ5 Fluorescent Probe is compatible with many existing protocols across a wide range of instrumentation platforms.

Fluorescence cell-based assays, such as flow cytometry, in-cell ELISA, fluorescence microscopy and high-content imaging require a fluorescent label to identify individual cells. When using multiple fluorescent probes to detect different cellular targets or activities, each probe must have a fluorescent spectrum different than the other probes. The blue-fluorescent DNA-binding probes, Hoechst and DAPI are frequently used; however, these probes cannot be used when UV illumination is unavailable or other blue-emitting fluorescent probes are used. Therefore, nuclear probes that emit in a color other than blue are useful for cell identification and counting, and for determining nuclear morphology and DNA content.

DRAQ5 Fluorescent Probe emits in the far-red region, is lipophilic and crosses cell and nuclear membranes in live and fixed cells and tissues for rapid DNA staining. This stain is water-soluble, supplied ready to use and does not require RNase, cell lysis, or a washing step, making it compatible with automation. Because DNA staining is stoichiometric, the DRAQ5 Fluorescent Probe can be used for DNA content analysis in cell proliferation studies. DRAQ5 Fluorescent Probe is usually added as the last stain in a labeling procedure because no washing is required. Alternatively, add this probe in assay medium for a live cell assay.

SYBR Safe DNA gel stain was developed specifically for reduced mutagenicity – to be safer than ethidium bromide for staining DNA in agarose or acrylamide gels. SYBR Safe stain is not only less mutagenic than ethidium bromide, but SYBR Safe stain's detection sensitivity is better than that of ethidium bromide. E-Gel® CloneWell™ Agarose Gels with SYBR® Safe DNA Gel Stain, 0.8% (Invitrogen™)

The E-Gel® CloneWell 0.8% SYBR® Safe gels and E-Gel® iBase™ Power System provide the easiest, most effective method for isolating DNA bands separated by agarose gel electrophoresis. You will:

How it Works
The E-Gel® CloneWell 0.8% SYBR® Safe gel features two rows of wells (Figure 1). To extract your DNA band of interest without conventional gel purification, simply load your sample into the top row and electrophorese until your band moves into the bottom row of wells. Then use a pipette to easily remove your purified DNA band. Guides on the gel cassette help you track your band of interest so you know when your DNA of interest is ready for retrieval. There is no need to cut a gel slice and risk losing your DNA during a gel purification step. The 0.8% agarose concentration is ideal for separating bands up to 5 kb in length. SYBR® Safe DNA Gel Stain is incorporated into the gel itself, allowing you to visually monitor migration on a blue-light transilluminator, such as the Safe Imager™ Blue-Light Transilluminator.*

Self-contained Power System
E-Gel® CloneWell SYBR® Safe gels run in the E-Gel® iBase™ Power System (the new E-Gel® iBase™ Power System is compatible with all low throughput E-Gels). This self-contained device includes a built-in power supply. Simply connect the AC adaptor provided and plug into an electrical outlet. Each E-Gel® agarose gel is ready for use with agarose, electrodes, and DNA stain (ethidium bromide or SYBR® Safe) packaged inside a dry, disposable, UV-transparent cassette. Safer than ethidium bromide
SYBR® Safe DNA Gel Stain has been specifically formulated and evaluated in a battery of toxicity tests with results indicating so that it is less mutagenic and safer for you to work with than ethidium bromide. An independent laboratory showed reduced mutagenicity of SYBR® Safe stain compared to ethidium bromide using the Ames test (see figure). You can further decrease your exposure risk by using visible blue-light illumination with SYBR® Safe stain rather than UV illumination. This is especially valuable when performing exposure-intensive protocols like cutting bands out of gels.

Excellent sensitivity
SYBR® Safe stain offers excellent sensitivity in nucleic acid visualization and documentation applications with either UV excitation or blue-light excitation. When bound to nucleic acids, the green-fluorescent SYBR® Safe stain has fluorescence excitation maxima at ~280 and ~502 nm, and an emission maximum at ~530 nm (see figure). Plus, when used with blue light illumination, SYBR® Safe stain has less background fluorescence than ethidium bromide–stained gels illuminated with UV light.

Easy to use
SYBR® Safe stain is supplied as a 10,000X concentrate in DMSO which can be used just like a solution of ethidium bromide. SYBR® Safe stain can be mixed into an agarose gel for staining during electrophoresis or the gel can be incubated in a solution of SYBR® Safe stain following electrophoresis. E-Gel® 96 Agarose Gels are bufferless, pre-cast gels designed for fast, high-throughput DNA electrophoresis. Each gel contains 96 sample lanes and 8 marker lanes that are ideal for screening multiple PCR products, plasmid preparations, and restriction digests. The unique 96-well staggered-well format provides a 1.6 cm run length that resolves between 100 bp and 10 kb. SYTOX Orange dye stains nucleic acids in cells with compromised membranes. This stain is useful as an indicator of cell death and is much brighter than propidium iodide (P-1304).
