In this course we will walk you through a basic CRISPR cell engineering workflow, from design and synthesis of target specific guide RNAs (gRNAs) to analysis of gene editing efficiency. This introductory workshop is structured as a combination of lectures and discussions with a hands-on laboratory instruction. Our experienced trainers will guide you through generating a knockout and knockin construct using CRISPR-Cas9 and will work closely with you to help ensure you are successful in learning these new methods.

Workshop agenda topics include:

  • gRNA synthesis using PCR and in vitro transcription (IVT) 
  • Delivery of IVT gRNAs and Cas9 mRNA into a human cell line using a lipid-based transfection reagent, and delivery of gRNA and Cas9 protein into the cell line using electroporation to generate a gene knockout
  • Introduction of an SNP change in a fluorescent protein through homology directed repair (HDR) with a single-stranded DNA oligonucleotide donor template
  • Determination of general editing efficiency with a PCR-based DNA mismatch method (genomic cleavage detection assay) and detection of the SNP change through analysis of fluorescent protein expression
