Sample Separation for Protein Analysis
Streamline your workflows with our sophisticated separation solutions
Nano LC systems are designed to optimize low flow separations and facilitate easy coupling to mass spectrometry to provide the best resolution, sensitivity, and selectivity for nano LC and proteomics and clinical translational biomarker discovery applications. We have pioneered the field of nano HPLC, a technique that generally involves the application of columns with an internal diameter of 75 µm and low flow rates of around 300 nL/min. We offer several solutions to accommodate reliable and robust nano LC as a front end to mass spectrometry and to fulfill three typical requirements.

High pressure liquid chromatography (HPLC)
Most single channel HPLC systems are comprised of modules connected by stainless steel tubing. The flexibility to select different styles of autosamplers, pumps and detectors is important to most laboratories since their needs vary based on the applications they plan to run. For clinical translational research, most laboratories seek high performance systems to meet their demanding needs in addition to support the connection to their mass spectrometer.
Nano LC systems are designed to optimize low flow separations and facilitate easy coupling to mass spectrometry to provide the best resolution, sensitivity, and selectivity for nano LC and proteomics and clinical translational biomarker discovery applications. Dionex has played a pioneering role in the field of nano HPLC, a technique that generally involves the application of columns with an internal diameter of 75 µm and low flow rates of around 300 nL/min. These configurations are ideal for analysis with limited sample starting amounts, due to the sensitivity enhancement of the smaller column i.d. We offer several solutions to accommodate reliable and robust nano LC as a front end to mass spectrometry and to fulfill three typical requirements:
- High sensitivity to allow detection of femtomoles or even lower
- Maximum robustness and repeatability for consistent results at a global level
- Increased resolution and selectivity for more reliable identification
Next Generation UHPLC Technologies: Change the Landscape in LC
Mike Oliver and Rainer Bauder discuss how next generation uHPLC and solid core column separation technology can interrogate more sample sets faster and more effectively.
For high-resolution and high-sensitivity separations that are ideally suited for use with mass spectrometry and designed for laboratories with less throughput or where sample preparation is done as a separate step in their analytical workflow. To ensure high separation efficiency, low gradient response times and superior mixing performance we offer perfectly matched HPLC and UHPLC system components.
Multi channel LC separation and online extraction
Turbulent flow liquid chromatography was developed in the late 1990s, and combines 'size exclusion' and traditional stationary phase column chemistry to separate macromolecules, such as proteins, from smaller molecules and analytes of interest in biological fluids. The process is very rapid, and the instrumentation and software have been developed for fully automated, on-line extraction of neat biological fluids.
With TurboFlow™ Technology, dramatically increase the return on your capital investment in mass spectrometer technology by improving data quality, lowering per-sample costs and increasing sample throughput using automated online sample preparation and multichannel liquid chromatography multiplexing.
High-throughput Bioanalytical LCMS Methods for Clinical Research and Drug Monitoring Research
Lewis Couchman and David Taylor from King’s College Hospital in London discuss potential application improvements from deploying turboflow technology.
Multi-channel intelligent operation: maximize productivity
Achieve up to four times throughput on your single triple quadrupole or high resolution mass spectrometer using the multi-channel turbulent flow liquid chromatography system. Enable up to four unique methods or maximize a single method using the available channels.
Because the system enables you to operate with or without on-line extraction, you have the flexibility to do offline or automated on-line extraction regardless of your laboratory workflow design.
Online extraction using size exclusion and selectivity
On line extraction sample cleanup is accomplished using TurboFlow Technology and is ideal for reproducibly separating analytes from complex, dirty matrices such as plasma and serum. This technique is able to efficiently remove interferences that can cause ion suppression in mass spectrometry which subsequently significantly reducing signal.
The unique features of our systems allow the compounds of interest to be retained on the column while the plasma or other biological matrices quickly and completely elutes through to waste. Eliminate the need for lengthy sample preparation steps, such as solid phase extraction (SPE) and protein precipitation (PPT). TurboFlow technology minimizes ion suppression for cleaner sample extracts, which is a significant advantage compared to SPE and PPT. As a result, it is the preferred method for cleaning biological fluids.
