Immunofluorescent staining of PC12 cells using PA5-19783, anti-160 kD Neurofilament Medium antibody. The cells were fixed with PFA (4%) for 10 minutes, permabilised with BSA(1%), normal goat serum (10%) and glycine (0.3 M) in 0.1% T-BST for 1 hour and exposed to the primary antibody at a concentration of 1 ug/ml overnight at 4C. The secondary antibody was a 448 fluorescence conjugated Goat anti-rabbit IgG (green) at a dilution of 1:1000. A WGA- 594 fluorescent conjugated stain was used to label plasma membranes (red) and the nuclei stain was DAPI (blue).

Neurofilaments are type IV intermediate filament heteropolymers composed of light, medium, and heavy chains. Neurofilaments comprise the axoskeleton and functionally maintain neuronal caliber. They may also play a role in intracellular transport to axons and dendrites. This gene encodes the medium neurofilament protein. This protein is commonly used as a biomarker of neuronal damage. Alternative splicing results in multiple transcript variants encoding distinct isoforms.
