Western blot detection of AP3 in 20µg HeLa whole cell lysate. Samples were separated on a SDS-PAGE, transferred onto nitrocellulose, and probed using an AP3 polyclonal antibody (Product # PA1-31774) at a dilution of 1:500 and detected using a 1:5000 dilution of HRP-labeled Rabbit anti-Goat IgG for 1 hour at room temperature. Chemiluminescence was used for signal detection at a 30-sec exposure time. The lower molecular weight band most likely represents non-specific binding.

AP3D1 is a subunit of the AP3 adaptor-like complex, which is not associated with clathrin. The AP3D1 subunit is implicated in intracellular biogenesis and trafficking of pigment granules and possibly platelet dense granules and neurotransmitter vesicles.
