IImmunofluorescence analysis for detection of Human Serum Albumin was carried out in 70% confluent log phase Hep G2 cells. The cells were fixed with methanol for 5 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with Human Serum Albumin Rabbit Polyclonal Antibody, FITC conjugate (Product # PA5-16600) at a dilution of 1:250 in 0.1% BSA and incubated for 3 hours at room temperature. Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). Cytoskeleton was stained with alpha-tubulin Monoclonal Antibody (Product # 322500, 1µg/ml) followed by Goat anti-Mouse IgG Secondary Antibody, Alexa Fluor® 594 conjugate (Product # A11032, 1:400). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (S36938). Panel d represents the composite image showing cytoplasmic localization of Human Serum Albumin. Panel e shows the no primary antibody control. The images were captured at 60X magnification.

The PA5-16600 immunogen is albumin isolated from human serum.
Albumin is a soluble, monomeric protein which comprises about one-half of the blood serum protein. Albumin functions primarily as a carrier protein for steroids, fatty acids, and thyroid hormones and plays a role in stabilizing extracellular fluid volume. Albumin is a globular unglycosylated serum protein of molecular weight 65,000. Albumin is synthesized in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum. The product, proalbumin, is in turn cleaved in the Golgi vesicles to produce the secreted albumin.
