Aldolase A Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemistry/Immunofluorescence analysis of Aldolase-A in SK-Br-3 cells (green). Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA for 15 minutes at room temperature. Cells were probed with Aldolase A Recombinant Monoclonal Antibody (Product # MA5-41240) at a dilution of 1:50 for 1 hour at room temperature, washed with PBS. Alexa Fluor®488 Goat anti-Rabbit IgG was used as the secondary antibody at 1:1,000 dilution. The nuclear counter stain is DAPI (blue).

Aldolase A Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry (Paraffin) analysis of Aldolase-A in paraffin-embedded human fetal skeletal muscle tissue. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with Aldolase A Recombinant Monoclonal Antibody (Product # MA5-41240) at a dilution of 1:50 for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Aldolase A Antibody in Western Blot (WB) — Western Blot analysis of Aldolase-A using Aldolase A Recombinant Monoclonal Antibody (Product # MA5-41240) at a dilution of 1:500 was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG-HRP Secondary Antibody at 1:200,000 dilution was used for 1 hour at room temperature. Lane 1: A549 cell lysate, Lane 2: Hela cell lysate, Lane 3: Mouse liver tissue lysate, Lane 4: Rat liver tissue lysate, Lane 5: Rat spleen tissue lysate. .

Target Information

Aldolase encodes a member of the class I fructose-bisphosphate aldolase protein family. The encoded protein is a glycolytic enzyme that catalyzes the reversible conversion of fructose-1,6-bisphosphate to glyceraldehyde 3-phosphate and dihydroxyacetone phosphate. Three aldolase isozymes (A, B, and C), encoded by three different genes, are differentially expressed during development. Mutations in this gene have been associated with Glycogen Storage Disease XII, an autosomal recessive disorder associated with hemolytic anemia. Disruption of this gene also plays a role in the progression of multiple types of cancers. Related pseudogenes have been identified on chromosomes 3 and 10.
