Immunofluorescent analysis of Alpha methylacyl-CoA-racemase (green) showing staining in the in the peroxisomes and mitochondria of human prostate carcinoma tissue (right) compared to a negative control without primary antibody (left). Formalin-fixed tissue was permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Tissue was probed with an Alpha methylacyl-CoA-racemase monoclonal antibody (Product # MA5-14576) in 3% BSA-PBS at a dilution of 1:20 and incubated overnight at 4°C in a humidified chamber. Tissue was washed with PBST and incubated with a DyLight-conjugated secondary antibody in PBS at room temperature in the dark. Nuclei (blue) were stained with Hoechst or DAPI. Images were taken at a magnification of 60x.

Alpha methylacyl-CoA-racemase (AMACR), also known as p504s is an enzyme involved in the metabolism of branched-chain fatty acids. p504s is expressed in various normal tissues and carcinomas, while it is overexpressed over 30-fold in prostate carcinoma compared to benign glandular tissues1. When p504s is overexpressed in atypical glands without basal cells, it establishes a diagnosis of prostatic carcinoma. High-grade prostatic intraepithelial neoplasia, atypical adenomatous hyperplasia and urothelial carcinoma may also express p504s.
