Annexin V Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemistry analysis of Annexin V using anti-Annexin V antibody (Product # PA5-78784) . Annexin V was detected in a section of U2OS cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 mins. The cells were blocked with 10% goat serum and then incubated with 2μg/mL rabbit anti-Annexin V antibody (Product # PA5-78784) overnight at 4°C. DyLight®488 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

Annexin V Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of Annexin V on paraffin-embedded human intestinal cancer tissue. Sample was incubated with Annexin V polyclonal antibody (Product# PA5-78784).

Annexin V Antibody in Immunohistochemistry (Frozen) (IHC (F)) — Immunohistochemical analysis of Annexin V in frozen section of human placenta tissues. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/mL rabbit anti-Annexin V antibody (Product # PA5-78784) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.

Annexin V Antibody in Immunohistochemistry (IHC) — Immunohistochemical analysis of Annexin V using anti-Annexin V antibody (Product # PA5-78784) Annexin V was detected in paraffin-embedded section of human placenta cancer tissues. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution ) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2μg/mL rabbit anti-Annexin V antibody (Product # PA5-78784) overnight at 4°C. FITC Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

Annexin V Antibody in Western Blot (WB) — Western blot was performed using anti-Annexin V Polyclonal Antibody (Product # PA5-78784) and a 35 kDa band corresponding to Annexin V was observed across the cell lines and tissue tested. Whole cell extracts (30 µg) of HeLa (Lane 1), Jurkat (Lane 2), MCF7 (Lane 3), MDA-MB-231 (Lane 4), Hep G2 (Lane 5) and Mouse Liver (Lane 6) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.5 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Annexin V Antibody in Flow Cytometry (Flow) — Flow Cytometry of Annexin V in SiHa cells (blue line), isotype control rabbit IgG (green line) and unlabeled (red line). Samples were blocked with 10% goat serum, incubated with Annexin V Polyclonal Antibody (Product # PA5-78784) at a dilution of 1 μg (per 1x10^6 cells), followed by DyLight®488 conjugated goat anti-rabbit IgG (for 30 minutes at 20°C) using 5-10 μg (per 1x10^6 cells) dilution.

Annexin V Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. HeLa cells were transfected with Annexin V siRNA and decrease in signal intensity was observed in western blot application using Anti-Annexin V Polyclonal Antibody (Product # PA5-78784).

Target Information

The protein encoded by this gene belongs to the annexin family of calcium-dependent phospholipid binding proteins some of which have been implicated in membrane-related events along exocytotic and endocytotic pathways. Annexin 5 is a phospholipase A2 and protein kinase C inhibitory protein with calcium channel activity and a potential role in cellular signal transduction, inflammation, growth and differentiation. Annexin 5 has also been described as placental anticoagulant protein I, vascular anticoagulant-alpha, endonexin II, lipocortin V, placental protein 4 and anchorin CII. The gene spans 29 kb containing 13 exons, and encodes a single transcript of approximately 1.6 kb and a protein product with a molecular weight of about 35 kDa.
