Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is liver tissue.
The Annexin family of calcium-binding proteins is composed of al least ten mammalian genes. It is characterized by a conserved core domain which binds to phospholipids in a Ca2+-dependent manner and a unique amino terminal region which may confer binding specificity. The Annexin family has been implicated as regulators of such diverse processes as ion-flux, endocytosis and exocytosis, and cellular adhesion. When overexpressed in A431 cells, Annexin VI causes a partial reversal of the transformed phenotype.
