Western blot analysis was performed on 10ul Hep G2 conditioned media (Lane 1) and Human Plasma (Lane 2). The blot was probed with Anti-ApoA1 Rabbit Oligoclonal Antibody (Product# 710263, 2 ug/ml) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 ug/ml, 1:2500 dilution). A 28 kDa band corresponding to ApoA1 was observed in both lanes tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 12 % Bis-Tris gel (Product # NP0342BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody. Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Intact IgG appears on a non-reducing gel as ~150 kDa band and upon reduction generating a ~25 kDa light chain band and a ~50 kDa heavy chain.
Apo A1 (Apolipoprotein A1) is synthesized in the liver and small intestine, and is regulated by the dietary intake of lipids. It is the major constituent of human high density lipoproteins (HDL) and plays an important role in metabolic and transport function of lipoproteins of the serum. It is regulated by the dietary intake of lipids, and acts as an activator for the enzyme lecithin cholesterol acyl transferase which converts cholesterol to cholesteryl esters. During lypolysis in the peripheral tissue, Apo A1 can also play a role as an acceptor for surface components of very low density lipoproteins and chylomicrons. The gene encoding Apo A1 is found on chromosome 11.
