Western blot analysis of Bax was performed by loading 25 ug of MCF-7 (Lane 1) and L929 (Lane 2) cell lysates onto an SDS polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked at 4°C overnight. The membrane was probed with a Bax monoclonal antibody (Product # MA5-14003) at a dilution of 1:20 overnight at 4°C, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hr at room temperature in the dark. Chemiluminescent detection was performed using Pierce ECL Plus Western Blotting Substrate (Product # 32132). Results show a band at approx. 21 kDa.

MA5-14000 targets Bax in ICC/IF, FACS, IHC (P) and WB applications and shows reactivity with Human, mouse, and Rat samples.
The MA5-14000 immunogen is a Synthetic peptide, aa 12-24 (Cys-GPTSSEQIMKTGA), of human Bax protein. This amino acid sequence is shared by human, mouse and rat bax protein.
Bcl-2, Bax, and Bcl-X are members of a family of proteins that regulate apoptosis. In contrast to Bcl-2alpha and Bcl-XL, overexpression of bax protein accelerates the programmed cell death (apoptosis).
