Western blot analysis was performed on human plasma (5 ul) (Lane 1). The blots were probed with Mouse Anti-C1 Inhibitor Monoclonal Antibody (Product # LFMA0136, 0.5 ug/ml) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 ug/ml, 1:4000 dilution). A 104 kDa band corresponding to C1 Inhibitor was observed across the samples tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

C1 inhibitor (C1-INH) is a serine protease inhibitor that inactivates several different proteases in the complement, contact, coagulation, and fibrinolytic systems. Though C1-INH has 478 residues (53kDa), the circulating protein is approximately 104 kDa by post-translational modification. Deficiency of C1-INH results in hereditary angioedema. In addition, C1-INH may prove useful in a variety of other diseases including septic shock, reperfusion injury, hyperacute transplant rejection, traumatic and hemorrhagic shock.
