CSNK1A1 (casein kinase 1 alpha 1) and CSNK1A1L (casein kinase 1 alpha 1-like) are serine/threonine protein kinases involved in various cellular processes. CSNK1A1 is located on chromosome 5 (5q32) and functions as a putative tumor suppressor, playing a role in cell cycle regulation, apoptosis, and autophagy. It is involved in the CGAS-STING pathway, inhibiting the overactivation of type I interferon signaling by promoting autophagic degradation of STING1. Additionally, CSNK1A1 has implications in certain cancers and myelodysplastic syndromes where its regulation affects cell proliferation and sensitivity to therapies. In contrast, CSNK1A1L is a paralog of CSNK1A1 that shares similar functional domains and potentially overlapping functions, although its specific role and regulatory mechanisms remain less characterized. Both genes are involved in critical pathways regulating cell survival and immune response, and their dysregulation is associated with disease states.
