CXCL2 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of C-X-C motif chemokine 2 (CXCL2) was performed using 70% confluent log phase untreated and LPS treated (100 ng/mL for 7 hr) RAW 264.7 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with CXCL2 Monoclonal Antibody (40605) (Product # MA5-23737) at 1:100 in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32790), (1:2500), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b:Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing staining for CXCL2 in golgi-like structures. Panel e represents untreated RAW 264.7 cells with no staining for CXCL2. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification in EVOS™ M7000 Imaging System (Product # AMF7000).

CXCL2 Antibody in Neutralization (Neu) — Neutralization assay of recombinant mouse CXCL2/MIP-2 chemoattracts the BaF3 mouse pro-B cell line transfected with human CXCR2 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin. Chemotaxis elicited by recombinant mouse CXCL2/MIP-2 (2 ng/mL) is neutralized (green line) by increasing concentrations of mouse CXCL2/MIP-2 Monoclonal Antibody (Product # MA5-23737). The ND50 is typically 1-4 µg/mL.

CXCL2 Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. RAW 264.7 cells were transfected with C-X-C motif chemokine 2 siRNA and decrease in signal intensity was observed in ICC application using Anti-CXCL2 Monoclonal Antibody (40605) (Product # MA5-23737). {KD}

Target Information
