CaMKIV Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of HepG2 cells using CAMK4 monoclonal antibody (Product # MA5-17038) (Green). Blue: DRAQ5 fluorescent DNA dye. Red: actin filaments have been labeled with phalloidin.

CaMKIV Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of paraffin-embedded ovarian cancer tissues using CAMK4 monoclonal antibody (Product # MA5-17038) followed with DAB staining.

CaMKIV Antibody in Western Blot (WB) — Knockdown of CaMKIV was achieved by transfecting SHSY5Y with CaMKIV specific siRNAs (Silencer® select Product # s2355). Western blot analysis (Fig. a) was performed using whole cell extracts from the CaMKIV knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with CaMKIV Monoclonal Antibody (8C5B8) (Product # MA5-17038, 1:1000 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Reduction of signal upon siRNA mediated knock down confirms that antibody is specific to CaMKIV.

CaMKIV Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. SH-SY5Y cells were transfected with CaMKIV siRNA and reduction in signal was observed in Western Blot using CaMKIV Monoclonal Antibody (8C5B8) (Product # MA5-17038).

MA5-17038 detects CAMK4 which has a predicted molecular weight of approximately 58kDa.

Target Information

CaM kinase IV (CaMKIV), in a calcium and calmodulin dependent manner, is implicated in phosphorylating numerous transcription factors including the CCAAT enhancer binding protein (C/EBP), serum response factor (SRF), and cyclic AMP response element binding protein (CBP). The nuclear localization of this protein is consistent with its role in mediating calcium-dependent gene expression. CaMKIV is particularly abundant in testis, T-cells, and neurons but is also found in other tissues to varying degrees. In neurons, CaMKIV is thought to play an important role in synaptic plasticity via its gene regulatory effects. In T-cells, this protein plays an important role in calcium signaling which could affect the transcription regulatory protein, nuclear factor of activated T-cells (NFAT). CaMKIV is encoded, along with calspermin, by the CaMKIV gene. It has been found that, in testes, CaMKIV is expressed in germ cells and found to be associated with chromatin. The association of CaMKIV with chromatin suggests a potential role in chromatin remodeling during nuclear condensation in spermatids.
