Calreticulin 3 (CALR3) is a gene located on chromosome 19p13.11, encoding a member of the calreticulin family of calcium-binding chaperones. Unlike the more ubiquitously expressed calreticulin (CALR), CALR3 is specifically expressed in the testes and plays a crucial role in spermatogenesis. CALR3 is involved in the quality control of proteins within the endoplasmic reticulum (ER) and assists in the folding and assembly of newly synthesized glycoproteins. This protein's function is essential for the maturation of sperm cells and male fertility, as it helps ensure the proper formation and function of sperm-specific proteins. Mutations in CALR3 have been associated with familial hypertrophic cardiomyopathy, reflecting its potential involvement in cardiac muscle function through mechanisms that remain to be fully elucidated. Research continues to explore CALR3's specific roles in reproductive biology and its implications in hereditary conditions.
