Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human Cardiotrophin-1. Anti-Human Cardiotrophin-1-specific antibody was purified by affinity chromatography employing an immobilized Human Cardiotrophin-1 matrix.
Sandwich ELISA: To detect Human Cardiotrophin-1 by sandwich ELISA (using 100 µL/well antibody solution) a concentration of 0.5-2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human Cardiotrophin-1 (500-P101Bt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human Cardiotrophin-1.
Western Blot: To detect Human Cardiotrophin-1 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant Human Cardiotrophin-1 is 1.5-3.0 ng/lane, under reducing or non-reducing conditions.
Cardiotrophin-1 (CT-1) is a member of the IL-6 family of cytokines, which signal through gp130 receptor complexes. gp130 complexes with several different receptor subunits to transmit signals from Cardiotrophin-1, IL-6, LIF, OSM, CNTF and IL-11. Cardiotrophin-1 binds to and activates the leukemia inhibitory factor (LIF) receptor/gp130 receptor complex and has been shown to induce hypertrophy in cardiac myocytes in vitro. Cardiotrophin-1, a secreted protein expressed at high levels in myocardium during cardiogenesis, has been shown to promote proliferation and survival of embryonic cardiomyocytes, suggesting a role for Cardiotrophin-1 in the activation of gp130 during cardiac development.
