Immunofluorescent analysis of Cdk2 (green) in HeLa cells. The cells were fixed with formalin for 15 minutes, permeabilized with 0.1% Triton X-100 in TBS for 10 minutes, and blocked with 5% Normal Goat Serum (Product # 31872) for 15 minutes at room temperature. Cells were stained with or without Cdk2 monoclonal antibody (Product # MA5-13725), at a dilution of 1:50 overnight at 4C, and then incubated with a DyLight 488 goat anti-mouse IgG secondary antibody (Product # 35503) at a dilution of 1:1000 for 30 minutes at room temperature (both panels, green). Nuclei (both panels, blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken on a Thermo Scientific ToxInsight at 20X magnification.

p33cdk2 associates with cyclin A in human cells. Kinase activity associated with cyclin A-cdc2 is found only in G2-phase. Cdk2 also complexes with cyclins E, D1, and D3. Cyclin E-cdk2 kinase is active in the G1- and S-phases of the cell cycle and is important (as does cyclin A-cdk2) for the progression from G1-to S-phase. The levels of cyclin A-cdk2 is maximal at the G1/S transition and both cdk2 and cyclin A are found associated with DNA in the initiation complex during replication. Rb protein acts as substrate for cdk2-cyclin E and/or cdk2-cyclin A in vivo. Cdk2 is activated and specifically localized to the nucleus during late G1-, S-, and G2-phase.
