Immunohistochemical analysis of Collagen III in formalin-fixed, paraffin-embedded liver tissue using a Collagen III polyclonal antibody (Product # PA1-28870) at a dilution of 1:500 for 4°C for 24 hrs and detected using a Peroxidase biotin-streptavidin rabbit secondary antibody at a dilution of 1:10,000 for 45 min at room temperature. A: Central Vein (CV) fibrosis, B: Non-fibrotic CV, C: Perisinusodial fibrosis, D: Non-fibrotic area, E: Protat tract fibrosis, F: Septal fibrosis (arrow). Antigen retrieval was not required. Collagen type I is intra and extracellular. Staining: 3.3"e;-diaminobenzidine tetrahydrochloride was used as the chromogen. Nuclei were counterstained purple with hematoxylin.

PA1-28870 has been successfully used in ELISA, immunohistochemistry (frozen tissue), immunoprecipitation, and Western blot applications. NOTE: The preparation of a specific antibody to Collagen III's uninterrupted ""Glycine-X-Y"" triplet repeat requires a non-denatured epitope format. Consequently, it is recommended to use this antibody in a Western blot using non-denaturing or "native" conditions.
The PA1-28870 immunogen is Collagen Type III from human and bovine placenta.
Store at 4°C short term. For extended storage add 50% glycerol, aliquot and store at -20°C, avoiding freeze/thaw cycles.
Type III collagen is present in the walls of arteries, intestines and the uterus, where it strenghtens the connective tissue.
