Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is colon carcinoma.
The cripto gene is expressed both in embryonic stem cells and during the early phases of embryo development, while in adults it is reactivated in a wide range of epithelial cancers. The expression of Cripto-1 may represent a useful marker to identify cancer stem cells in melanoma, and possibly other aggressive tumors as well. It has been suggested that caveolin-1, a membrane protein that acts as a tumor suppressor in the mammary gland is down-regulated during the process of Cripto- 1 mediated mammary tumorigenesis.
