Immunofluorescent analysis of Cytohesin 2 in NIH-3T3 Cells. Cells were grown on chamber slides and fixed with formaldehyde prior to staining. Cells were probed without (control) or with a Cytohesin 2 monoclonal antibody (Product # MA1-061) at a dilution of 1:100 overnight at 4 C, washed with PBS and incubated with a DyLight-488 conjugated secondary antibody (Product # 35503). Cytohesin 2 staining (green), F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue) is shown. Images were taken at 60X magnification.

MA1-061 detects recombinant human and mouse cytohesin-2 (ARNO).
MA1-061 has been successfully used in IHC (P), immunoprecipitation and IF procedures. By immunoprecipitation this antibody efficiently precipitates GFP-labeled ARNO. MA1-061 does not work in Western blot procedures.
The MA1-061 immunogen is purified, His-tagged cytohesin-2.
The protein encoded by this gene is a member of the PSCD family. Members of this family have identical structural organization that consists of an N-terminal coiled-coil motif, a central Sec7 domain, and a C-terminal pleckstrin homology (PH) domain. The coiled-coil motif is involved in homodimerization, the Sec7 domain contains guanine-nucleotide exchange protein (GEP) activity, and the PH domain interacts with phospholipids and is responsible for association of PSCDs with membranes. Members of this family appear to mediate the regulation of protein sorting and membrane trafficking. The encoded protein exhibits GEP activity in vitro with ARF1, ARF3, and ARF6 and is 83% homologous to CYTH1. Two transcript variants encoding different isoforms have been found for this gene.
