DNA-PK is a trimeric nuclear protein that contains the DNA-dependent protein kinase catalytic subunit (DNA-PKcs) and the Ku heterodimer. These proteins are required for the repair of DNA double-strand breaks and V(D)J recombination. DNA-PKcs is a 460kDa serine-threonine kinase and is a member of the phosphatidylinositol (PI) 3-kinase family of proteins and most similar to the subset of proteins involved in cell cycle checkpoint regulation and DNA damage sensing, such as, ATM (ataxia-telangiectasia mutated) protein.
