Immunohistochemistry was performed on 4% paraformaldehyde-fixed frozen mouse retina tissue sections. Tissues were blocked in 3% normal goat serum + 0.3% Triton X-100 for 1 hour at room temperature and probed with a Degenerative Spermatocyte Homolog 1 (DSH1) polyclonal antibody (Product # PA1-12912) at a dilution of 1:500 overnight at 4°C . Tissues were washed extensively with PBS. Detection was performed using a Cy3-conjugated goat anti-rabbit IgG secondary antibody at a dilution of 1:500. The product of the DEGS1 gene is a member of the membrane fatty acid (sphingolipid delta 4 ) desaturase family. Reported to function by inserting double bonds at delta 4 position in fatty acids. DEGS1 is an intracellular enzyme. Overexpression of this gene inhibited biosynthesis of the EGF receptor, suggesting a possible role of a fatty acid desaturase in regulation biosynthetic processing of the EGF receptor. Two splice variants have been identified.
