Human HDFn cells were paraformaldehyde fixed (4%, 20 min) and Triton X-100 permeabilized (0.1%, 15 min). The cells were incubated with the antibody at 1 µg/mL overnight at 4°C. Secondary antibody: (red) Alexa Fluor® 594 goat anti-mouse IgG (H+L) at a 1/1000 dilution for 1h. 10% Goat serum was used as the blocking agent for all blocking steps. DAPI was used to stain the cell nuclei (blue). Target protein locates mainly in mitochondria.

Inhibitors of apoptosis proteins (IAP's) block cell death by caspase inhibition. X linked inhibitor of apoptosis (XIAP) is a member of the IAP family. Smac/Diablo is expressed in a variety of tissues and has been identified as a negative regulator of XIAP. Smac/Diablo is a mitochondrial protein that enters the cytosol when cells undergo apoptosis, and moderates the caspase inhibition of IAPs.
