Immunofluorescent analysis of FKBP52 (green) in HeLa cells either left untreated (left panel) or treated with 0.5mM copper sulfate (right panel) for 24 hours. Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA (Product # 37525) for 15 minutes at room temperature. Cells were probed with a FKBP52 monoclonal antibody (Product # MA1-111) at a dilution of 1:1000 for at least 1 hour at room temperature, washed with PBS, and incubated with DyLight 488 goat anti-mouse IgG secondary antibody (Product # 35502) at a dilution of 1:400 for 30 minutes at room temperature. F-Actin (red) was stained with DyLight 554 Phalloidin (Product # 21834) and nuclei (blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken on a Thermo Scientific ArrayScan or ToxInsight Instrument at 20X magnification.

This affinity purified antibody detects an ~59 kDa protein corresponding to the apparent molecular mass of FKBP59 on SDS-PAGE immunoblots in samples from human, monkey, bovine, mink, rabbit, hamster and guinea pig (weakly) origins. An additional, unidentified 50kDa band is observed in some cell lysates. This antibody has been shown to block the ability of FK506 to promote neurite outgrowth from SH-SY5Y cells in a concentration-dependent manner between 50 and 100nM. MA1-111 has also been successfully used in immunohistochemistry (paraffin) applications.
This antibody was originally validated as part of a Thermo Scientific Cellomics High Content Screening Kit. The antibody sold separately may have slightly different performance and may need to be further optimized for the best results.
The protein encoded by this gene is a member of the immunophilin protein family, which play a role in immunoregulation and basic cellular processes involving protein folding and trafficking. This encoded protein is a cis-trans prolyl isomerase that binds to the immunosuppressants FK506 and rapamycin. It has high structural and functional similarity to FK506-binding protein 1A and thus may play a role in the intracellular trafficking of hetero-oligomeric forms of the steroid hormone receptors. This protein correlates strongly with adeno-associated virus type 2 vectors resulting in a significant increase in AAV-mediated transgene expression in human cell lines. Thus this encoded protein is thought to have important implications for the optimal use of AAV vectors in human gene therapy. The human genome contains several non-transcribed pseudogenes similar to this gene.
