Western blot analysis of Human Fractalkine was performed by loading 0.25µg of recombinant human Fractalkine onto a 4-20% Tris-HCl polyacrylamide gel. Protein was transferred to a PVDF membrane and blocked with 5% Milk/TBST for at least 1 hour. Membrane was probed with a human Fractalkine monoclonal antibody (Product # MA1-031) at a dilution of 1:1000 overnight at 4°C on a rocking platform. Membranes were washed in TBS-0.1% Tween-20 and probed with a goat anti-rabbit-HRP secondary antibody (Product # 31430) at a dilution of 1:10,000 for at least one hour. Membrane was washed and chemiluminescent detection performed using Super Signal West Dura (Product #34075).

Chemokine (C-X3-C motif) ligand 1 (CX3CL1) is a large cytokine protein of 373 amino acids, it contains multiple domains and is the only known member of the CX3C chemokine family. This molecule can exist in 2 forms: as membrane-anchored or as a shed 95-kD glycoprotein.
