Western blot analysis was performed on membrane enriched extracts (30 ug lysate) of HEK 293 (Lane 1), MCF7 (Lane 2) and Mouse Liver (Lane 3). The blot was probed with Anti-Fyn Mouse Monoclonal Antibody (Product # MA1-12607, 1-3 ug/ml) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.4 ug/ml, 1:2500 dilution). A 60 kDa band corresponding to Fyn was observed across the cell lines and tissue tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 10 % Bis-Tris gel (Product # NP0302BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody using iBind™ Flex Western Starter Kit (Product # SLF2000S). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

MA1-12607 detects Fyn in human, mouse and rat samples.
MA1-12607 has been successfully used in Western blot and immunohistochemistry procedures. Positive control of human or mouse T cell hyridomas, jurkat cells or T cells in tonsil suggested. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer pH 6.0 for 10-20 minutes followed by cooling at RT for 20 min.

Fyn is a membrane-associated non-receptor protein tyrosine kinase that belongs to the Src-family of kinases. The unique N-terminal domain of Fyn interacts with the CD3 and Delta chains of the TcR. p59 fyn can bind other proteins (p82 and p116) through its SH2 and SH3 domains, which may act as substrates or regulators of Fyn activity. Fyn is highly expressed in brain suggesting that it may have a role in the sensory nervous network and in myelination at early stages of CNS formation.
