Transcription factor HES-2, of the HES family of proteins, contains one basic helix-loop-helix (bHLH) domain and one orange domain. It is reported to express in vascular smooth muscle cells, and may have some role in their differentiation. Activation of Notch signaling pathway leads to activation of Hes family genes through the interaction between Notch intracellular domain and RBPSUH (CSL). Hes family proteins are mammalian homologs of Drosophila Hairy and Enhancer of split. The Hes family of transcriptional repressors are implicated in the cell fate determination of stem cells (or precursor cells) by suppressing the expression of tissue-specific transcriptional activators.
