Western blot analysis of HOXB4 in extracts of various cell lines. Samples were incubated with HOXB4 Monoclonal antibody (Product # MA5-37955) using a dilution of 1:1,000, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 180s.

Target Information
HOX homeobox genes play a role both in the early stem cell function as well as in later stages of hematopoietic differentiation. HOXB4, specifically, is a homeotic selector gene implicated in self-renewal of definitive HSCs. Expression of HOXB4 in primitive progenitors combined with culture on hematopoietic stroma induced a switch to the definitive HSC phenotype. Activation of WNT signaling in HSCs induced increased expression of HOXB4. Perturbations of HOX gene expression can cause leukemia.
