DB-1 exhibits high affinity binding for both rat and mouse natural and recombinant IFN-y and effectively neutralizes the antiviral activity of IFN-y in both mouse and rat. This antibody blocks induction of Ia antigen expression on murine keratinocytes.This antibody is suitable for in vivo and in vitro neutralizing studies. It does not react with rodent IFN-a, rodent IFN-b, or with human interferons.
Biological activity: 6000 neutralizing units/mg of protein. One unit is equivalent to the amount of antibody required to neutralize one laboratory unit of rat IFN-y antiviral activity, in which 6x10^6 units corresponds to 1 mg of pure rat interferon-y. For use in ELISA, this antibody is suitable as a coating antibody when diluted to 10 µg/mL and 100 µl per well.
Storage and reconstitution: store lyophilized antiobdy at or below 4°C. Reconstitute using 0.5 mL sterile distilled water. Upon reconstitution, apportion into working aliquots and store at or below -20°C, avoiding repeated freeze/thaw cycles. For long term storage, sodium azide should be added as a preservative.
IFN gamma, also called Type II interferon, macrophage activation factor, and immune interferon is produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, Staphylococcus enterotoxin B, phytohemaglutanin and other cytokines. IFN gamma is a glycoprotein that exists, functionally, as a homodimer of approximately 45 kDa. On SDS-PAGE it appears as a combination of 25, 20 and minor 15.5 kDa bands as a result of differential glycosylation. The biological activity of the IFN gamma homodimer is highly species specific and includes the following: antiviral activity, tumor antiproliferative activity, induction of class I and II MHC, macrophage activation, and enhanced immunoglobulin secretion by B lymphocytes. IFNg is involved in cytokine regulation and also acts synergistically with other cytokines.
