Sandwich ELISA analysis of mouse IL-10 was performed using a Mouse IL-10 Colorimetric ELISA kit (Product # EM2IL10) by loading 50 ul per well of Mouse IL-10 Recombinant Protein (Product # SMIL10) in triplicate at 3000, 1000, 333, 111, 37, and 0 pg/ml across a 1 ug/ml anti-mouse IL-10 (Product # MM011) pre-coated plate and incubating for 3 hours at room temperature. The plate was washed, then incubated with 50 ul per well of an anti-mouse IL-10 biotinylated antibody in triplicate at 1 ug/ml for 1 hour at room temperature. The plate was washed and incubated with 100 ul per well of Streptavidin-HRP (Product # N504) in all test wells at 1:40 for 30 minutes at room temperature. Detection was performed using 1-Step Ultra TMB substrate (Product # 34028) for 30 minutes at room temperature in the dark. The plate was then stopped with 0.16M sulfuric acid. Absorbances were read on a spectrophotometer at 450-550 nm.

MM011 detects IL-10 which has a predicted molecular weight of approximately 19 kDa.
This antibody is produced by injecting Rat IgG secreting hybridoma cells into the peritoneum of mice. The resulting ascites is collected from the mice and the antibody is purified.
This product is a Low Endotoxin formulation.
The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract.
