Normal human peripheral blood cells were unstimulated (left) or stimulated for 5 hours with the Cell Stimulation Cocktail (plus protein transport inhibitors) (Product # 00-4975) (right). Cells were stained intracellularly with Anti-Human CD4 APC (Product # 17-0049) and Anti-Human IL-13 PE using the Intracellular Fixation and Permeabilization Buffer Set (Product # 88-8824) and protocol. Cells in the lymphocyte gate were used for analysis.

Description: The 85BRD monoclonal antibody reacts with human interleukin-13 (IL-13). IL-13, also known as NC30, is a 12.5 kDa protein secreted by activated T helper cells, CD8+ T cells, and NK cells. The biological activities of IL-13 include suppression of macrophage cytotoxic activity, upregulation of IL-1RA expression and suppression of inflammatory cytokine expression.
Applications Reported: This 85BRD antibody has been reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested: This 85BRD antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells using the Intracellular Fixation and Permeabilization Buffer Set (cat. 88-8824) and protocol. Please refer to Best Protocols section (Protocol A: Two step protocol for (cytoplasmic) intracellular proteins). This can be used at 5 µL (0.125 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.

IL-13 is a cytokine involved in regulating inflammatory and immune responses. It down-regulates macrophage activity, thereby inhibiting the production of pro-inflammatory cytokines and chemokines, and is involved in allergen-induced asthma through mechanisms independent of IgE and eosinophils. IL-13 also regulates several stages of B cell maturation and differentiation. It up-regulates CD23 and MHC class II expression, and promotes IgE isotype switching of B cells. Structurally, IL-13 closely resembles IL-3, IL-4, IL-5 and GM-CSF.
