IL1RA Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence staining of Human IL1Ra in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 1% Triton X-100 in PBS, blocked with 10% serum, and incubated with IL1RA Monoclonal Antibody (2) (Product # MA5-29352, 1:60) at 37°C 1 hour. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-mouse IgG secondary antibody (green). Positive staining was localized to cytoplasm.

IL1RA Antibody in Western Blot (WB) — Western blot was performed using Anti-IL1RA Monoclonal Antibody (2) (Product # MA5-29352) and a 19 kDa band corresponding to Interleukin-1 receptor antagonist protein was observed across the cell lines tested. Whole cell extracts (30 µg lysate) of A-431 (Lane 1), SK-O-V3 (Lane 2) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:10,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).

IL1RA Antibody in Flow Cytometry (Flow) — Flow cytometric analysis of Human IL1RA expression on A431 cells. Cells were stained with IL1RA Monoclonal Antibody (2) (Product # MA5-29352), then a FITC-conjugated Secondary antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.

IL1RA Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. A-431 cells were transfected with Interleukin-1 receptor antagonist protein siRNA and decrease in signal intensity was observed in Western Blot application using Anti-IL1RA Monoclonal Antibody (2) (Product # MA5-29352). {KD}

Target Information

The protein encoded by this gene is a cytokine receptor that belongs to the interleukin 1 receptor family. This protein is a receptor for interleukin alpha (IL1A), interleukin beta (IL1B), and interleukin 1 receptor, type I(IL1R1/IL1RA). It is an important mediator involved in many cytokine induced immune and inflammatory responses. This gene along with interleukin 1 receptor, type II (IL1R2), interleukin 1 receptor-like 2 (IL1RL2), and interleukin 1 receptor-like 1 (IL1RL1) form a cytokine receptor gene cluster in a region mapped to chromosome 2q12.
