IL1RA Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence staining of Human IL1Ra in A431 cells. Cells were fixed with 4% PFA, permeabilzed with 1% Triton X-100 in PBS, blocked with 10% serum, and incubated with IL1RA Recombinant Rabbit Monoclonal Antibody (4) (Product # MA5-29353, 1:60) at 37°C 1 hour. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm.

IL1RA Antibody in Western Blot (WB) — Western blot was performed using Anti-IL1RA Recombinant Rabbit Monoclonal Antibody (Product # MA5-29353) and a 19 kDa band corresponding to Interleukin-1 receptor antagonist protein was observed across the cell lines tested. Whole cell extracts (30 µg lysate) of A-431 (Lane 1), SK-O-V3 (Lane 2) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

IL1RA Antibody in Flow Cytometry (Flow) — Flow cytometric analysis of Human IL1RA expression on A431 cells. Cells were stained with IL1RA Recombinant Rabbit Monoclonal Antibody (4) (Product # MA5-29353), then a FITC-conjugated Secondary antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.

IL1RA Antibody — Antibody specificity was demonstrated by siRNA mediated knockdown of target protein. A-431 cells were transfected with Interleukin-1 receptor antagonist protein siRNA and decrease in signal intensity was observed in Western Blot application using Anti-IL1RA Recombinant Rabbit Monoclonal Antibody (Product # MA5-29353). {KD}

Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. Target Information

The protein encoded by this gene is a cytokine receptor that belongs to the interleukin 1 receptor family. This protein is a receptor for interleukin alpha (IL1A), interleukin beta (IL1B), and interleukin 1 receptor, type I(IL1R1/IL1RA). It is an important mediator involved in many cytokine induced immune and inflammatory responses. This gene along with interleukin 1 receptor, type II (IL1R2), interleukin 1 receptor-like 2 (IL1RL2), and interleukin 1 receptor-like 1 (IL1RL1) form a cytokine receptor gene cluster in a region mapped to chromosome 2q12.
