Immunofluorescence analysis of KDM2A in Hela cells or 293T cells using Anti-KDM2A Recombinant Human Biotinylated Fab (730088). Hela cells or 293T cells were fixed with 3% Formaldehyde and lysed with 1% TritonX-100. Cells were then labeled with Anti-KDM2A Recombinant Human Biotinylated Fab (730088) and detected with Goat-anti-human Alexa Fluor dye F488 (green). FLAG Cells were double labeled with human Fab and detected with Goat-anti-human Alexa Fluor dye F488 (green) as well as with Mouse-anti-FLAG-M2 and detected with Goat-anti-Mouse Alexa Fluor dye F647 (yellow). The DNA in the nucleus was detected with DAPI (Blue). Control means cells were labeled only with secondary antibody Alexa Fluor Dye.

JHDM1a/FBXL11 is a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box),which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. JHDM1a/FBXL11 belongs to the Fbls class and, in addition to an F-box, contains at least 6 highly degenerated leucine-rich repeats.
