KLF2 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of Krueppel-like factor 2 was performed using 70% confluent log phase HUVEC cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with KLF2 Monoclonal Antibody (OTI3A10) (Product # MA5-26827) at 1:100 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Donkey anti-Mouse IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32766), (1:2000 dilution), for 45 minutes at room temperature (Panel a: Green) showing HUVEC cells treated with Rapamycin (2 µg/mL for 48hrs). Nuclei (Panel b:Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300 dilution). Panel d represents the merged image showing Nuclear localization for Rapamycin treated HUVEC cells. Panel e represents untreated cells. Panel f represents HUVEC cells treated with Recombinant human TNF Alpha (10 ng/mL for 4hrs). Panel g represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

KLF2 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry was performed on paraffin-embedded carcinoma of human kidney tissue. To expose target proteins, 1 mM EDTA in 10mM Tris, pH8.5, 120°C for 3min was used. Following antigen retrieval, tissues were probed with a KLF2 monoclonal antibody (Product # MA5-26827).

KLF2 Antibody in Western Blot (WB) — Western blot analysis of KLF2 in HEK293T cells in untransfected (Left lane) and transfected (Right lane) samples using 5 µg per lane. The samples were separated by SDS-PAGE and probed with KLF2 (Product # MA5-26827) monoclonal antibody.

KLF2 Antibody — Detection of altered subcellular localization of the target protein by cell treatment demonstrates antibody specificity. KLF2 belongs to the krueppel C2H2-type zinc-finger protein family. It contains 3 C2H2-type zinc fingers. KLF2 binds to the CACCC box in the beta-globin gene promoter and activates transcription.
