Western blot analysis of immunoglobulin (Ig) kappa light chain was performed by loading the indicated amounts of purified human IgG/kappa and 10ul of PageRuler Prestained Protein Ladder (Product # 26616) per well onto a 4-20% Tris-HCl polyacrylamide gel. Proteins were transferred to a PVDF membrane using the G2 Fast Blotter (Product # 62288), and blocked with 5% BSA in TBST for at least 1 hour at room temperature. Ig kappa light chain was detected at ~25kD using a biotinylated mouse anti-human Ig kappa light chain monoclonal antibody (Product # MA5-12114) diluted to a concentration of 0.5ug/ml in blocking buffer, overnight at 4C overnight on a rocking platform. The blot was washed in TBST, and probed with Streptavidin-HRP (Product # 21126) at a dilution of 1:40,000 for at least 30 minutes at room temperature. Chemiluminescent detection was performed using SuperSignal West Pico (Product # 34080). Note: IgG heavy chain (~55kD) was also detected when larger amounts of purified IgG were loaded per lane.

By Western blot, MA5-12114 detects Human Ig kappa light chain chain at ~25kD in human samples. At high concentrations, the antibody will also detect heavy chain.
By ELISA and WB, MA5-12114 does not react with human Ig lambda light chain.
Antibody to the kappa light chain of immunoglobulin is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.
