Western blot analysis of MMP10 was performed by loading 20 µg of Raji (lane1), HeLa (lane2), HEK-293 (lane3), Jurkat (lane4) and U2OS (lane5) cell lysates using Novex® NuPAGE® 4-12 % Bis-Tris gel (NP0321BOX), XCell SureLock™ Electrophoresis System (EI0002), Novex® Sharp Pre-Stained Protein Standard (LC5800), and iBlot® Dry Blotting System (IB21001). Proteins were transferred to a nitrocellulose membrane and blocked with 5 % skim milk for 1 hour at room temperature. MMP10 was detected at ~54 kDa using ABfinity™ MMP10 Recombinant Rabbit Monoclonal Antibody (701288) at 1-3 µg/ml dilution in 2.5 % skim milk at 4°C overnight on a rocking platform. Goat Anti-Rabbit IgG-HRP Secondary Antibody (G21234) at 1:5000 dilution was used and chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (WP20005).

MMP10 belongs to the matrix metalloproteinase family, and is generally secreted in its pro form, which is further modified by extracellular proteinases. As a metalloproteinase, MMP10 requires zinc and calcium as cofactors for activity. It is involved in the activation of pro-collagenase, catabolism of collagen, degradation of fibronectin, and tissue remodeling. In humans, the gene MMP10 located on chromosome 11.
