Western blot analysis was performed on nuclear-enriched extracts (30 ug lysate) of RSC-96 (Lane 1), T98G (Lane 2), MCF-7 (Lane 3), A549 (Lane 4), HCT 116 (Lane 5), K-562 (Lane 6), DU 145 (Lane 7), and PC-3 (Lane 8). The blots were probed with Rabbit Anti-MeCP2 Polyclonal Antibody (Product # PA1-888, 0.5 ug/ml) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 ug/ml, 1:4000 dilution). A 56 kDa band corresponding to MeCP2 was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

PA1-888 detects methyl CpG binding protein 2 (MeCP2) from human, mouse and rat cells and tissues.
PA1-888 has been successfully used in Western blot procedures. By Western blot, this antibody detects an ~56 kDa protein representing MeCP2 from AtT20 cell extract.
PA1-888 immunizing peptide corresponds to amino acid residues 469-486 from human MeCP2. This sequence is completely conserved in rat and mouse MeCP2. PA1-888 immunizing peptide (Cat. # PEP-121) is available for use in neutralization and control experiments.
