Mitochondria Antibody in Western Blot (WB) — Western blot of Mitochondria using Mitochondria Monoclonal Antibody (Product # MA5-12014) on HeLa Cells.

Mitochondria Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of Mitochondria was performed using 70% confluent log phase A549 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with Mitochondria Mouse Monoclonal Antibody, Biotin (Product # MA5-12014) at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Streptavidin, Alexa Fluor™ 488 conjugate (Product # S-11223) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). Mitochondria was also stained with MitoTracker® Red CMXRos (Product # M-7512). Panel d represents the merged image showing mitochondria co-localization. Panel e shows the no primary antibody control. The images were captured at 60X magnification.

Mitochondria Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Formalin-fixed, paraffin-embedded human liver stained with Mitochondria antibody using peroxidase-conjugate and AEC chromogen. Note granular staining of hepatocytes.

Product Specific Information

This antibody recognizes a 60kDa non-glycosylated protein component of mitochondria found in normal and malignant human cells.

Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min followed by cooling at room temperature for 20 min. Recommended positive controls: HeLa cells and liver tissue.

Target Information

Antibodies that recognize subcellular organelles or compartments of human cells are useful in identification of these organelles in cells, tissues, and biochemical preparations. Anti-mitochondria antibodies can be used to stain mitochondria in cell/tissue preparations and can be used as a marker in subcellular fractions.
