Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is prostate or prostate carcinoma.
The Nkx3.1 gene is an androgen regulated prostate-specific homeobox gene, located on chromosome 8p21. In mice the homologue Nkx3.1 is exclusively expressed in prostate epithelium. In humans, Nkx3.1 expression is also restricted to the prostate. Many data show that Nkx3.1 expression is highly, but not exclusively, specific for the prostate.
