Western blot analysis of Nanog was performed by loading 50ug of NCCIT and negative control HeLa cell lysates per well onto an SDS polyacrylamide gel. Proteins were transferred to a PVDF membrane. The membrane was blocked, and probed with an HRP-conjugated Nanog monoclonal antibody (Product # MA1-017-HRP) at a dilution of 1:1000 overnight at 4C. Chemiluminescent detection was performed using SuperSignal West Dura (Product # 34075).

Western blot analysis of MA1-017-HRP detects an ~38 kDa protein in embryonal carcinoma cells. MA1-017-HRP shows specificity to Nanog and is non-reactive to lysates from non-embryonal cell types (e.g. HeLa cell lysate).
